"We can beat anyone," said the 31 year-old, who is likely to play left-back in the absence of Danny Rose.
"We have shown that last year. Over two games we were the better team against Real Madrid [in the group stage last year] who were the eventual winners of the trophy.
"That is why it was a shame we could not do better last year because we felt we should have done better. That is why everyone has the feeling we can do it this year."
Due to playing in a less familiar full-back role, Vertonghen is likely to have to mark Dortmund's Jadon Sancho. The Englishman has been a revelation this season, and will be looking forward to an emotional homecoming in London.
Spurs are without Harry Kane and Dele Alli, who aren't expected to return from injuries until March, while Dortmund will be without influential forward Marco Reus.
