The Uruguayan limped off in the first half of Hull's victory against Southampton on Sunday and will be out for around a month.
Hull said via Twitter: "Abel Hernandez will be sidelined for around a month with the groin injury he picked up in the weekend win against Southampton".
Without last season's 22-goal man, Hull City will call on Dieumerci Mbokani in their next match against Sunderland as the former Norwich front man is the only available out-and-out striker to be available for the relegation crunch clash.
Hernandez's strike partner Will Keane picked up what appears to be a serious knee injury at the weekend while Norwegian Adama Diomande is also unavailable through suspension.
