Chris Wood headed a late winner after Sean Dyche's men twice came back from behind at Turf Moor.
Ollie Watkins opened the scoring for Villa but the hosts levelled through Ben Mee's header early in the second half.
Jack Grealish put the visitors back in front but Dwight McNeil levelled and Wood ensured Burnley won all three points.
Brighton were also in action against Fulham.
Champions Liverpool, who have taken just three points from their past five Premier League matches, are at Tottenham on Thursday.
