The Portuguese footballer continues to have discomfort in his right leg and it has now led to an injury after tests on Wednesday.
With three days left on Sunday, Joao Felix will not be able to be at the Santiago Bernabeu and will not play the derby that could leave Atletico out of Europe if they fall to Madrid and a series of results occur.
"Joao Felix suffers a muscle injury in his right leg produced in the match against Club Deportivo Leganés. Given the persistence of discomfort after that day, medical services have made tests to detect the injury, pending developments," said the club in its official statement.

Without Joao Felix, Atletico will have to rely on Alvaro Morata in the attack at the Santiago Bernabeu and will have to look for their partner at the top, a position for which the Argentinian Angel Correa or the Frenchman Thomas Lemar, who trained in that position in last Tuesday's training, will be in charge.
In addition to the most likely loss of the Portuguese playmaker, Atletico will face the Bernabeu without Diego Costa, Uruguayan Jose Maria Gimenez and Colombian Santiago Arias, all of whom are injured.
The red and whites will be looking to Koke Resurrección, who has been training separately in both sessions this week, and England's Kieran Trippier, who returned to work with the group on Wednesday.
